Penapisan Isolat Khamir Oleaginous dari Nektar Bunga dan Madu Hutan

Abstract


Khamir oleaginous mampu mengakumulasi lipid lebih dari 15% dari berat kering selnya. Pada kondisi nitrogen yang terbatas, khamir tersebut mampu mengakumulasi lipid hingga 70%. Lipid yang dihasilkan mengandung asam lemak seperti palmitat, oleat, dan linoleat, sehingga berpotensi untuk dimanfaatkan sebagai bahan baku produksi biodiesel. Khamir oleaginous umumnya hidup dilingkungan dengan konsentrasi karbon yang tinggi, antara lain nektar bunga dan madu. Penelitian ini bertujuan untuk melakukan penapisankhamiroleaginous yang diisolasi darinektar bunga dan madu hutan. Sampel nektar bunga diambil dari Kebun Raya Baturraden, Purwokerto,dan tempat pembibitan bunga di Kaliurang, Yogyakarta. Sedangkan sampel madu hutan diambil dari 11 kabupaten di Sulawesi. Uji kualitatif dilakukan dengan teknik pewarnaan sudan III dan uji kuantitatif dilakukan dengan mengukur berat lipid per berat kering biomassanya. Penelitian menghasilkan 6 isolat khamir oleaginous dari 47 isolat: PG12.2 (17,88%), PG12.3 (17,13%), PG5.4 (18,25%), AP1 (19,07%), PM2.1 (17,55%), dan PM3.2 (18,56%).Profil pertumbuhan isolat AP 1 menunjukkan bahwa kandungan lipid yang diproduksi masih menunjukkan peningkatan setelah penumbuhan selama 70 jam.
